BSL-3 laboratory design: facility and air features
A BSL-3 laboratory is a sealable room suite with controlled entry, directional airflow that cannot reverse on failure, exhaust that is not recirculated to the building, and surfaces that allow decontamination of the whole space. The BMBL, 6th edition (2020), lists these facility features, and your national rules and risk assessment set the exact values.

Scope of this guide
This guide covers building and air only. Biosafety levels are chosen by a risk assessment made by the owner. The WHO Laboratory biosafety manual, 4th edition (2020), is built on that risk-based approach. Pressure values, air change rates and leak-tightness limits differ by country, so none are given here. Read the current national code, such as China's GB 50346-2011 for the design, construction and acceptance of biosafety laboratories.

BMBL 6 facility features for BSL-3
Section IV, Biosafety Level 3, part D, secondary barriers.
| Feature | What the BMBL says | Design question |
|---|---|---|
| Entry | Separated from unrestricted traffic; two consecutive self-closing doors; change room or anteroom may sit between | Airlock layout and interlock |
| Handwash | Hands-free or automatic sink near the exit door; one in each zone | Sink positions |
| Surfaces | Seams, floors, walls and ceilings sealed; gaps around doors and vents sealable; floors seamless, sealed or poured with integral cove bases | Panel joints and sealants |
| Windows | All sealed | Glazing type |
| Ventilation | Ducted mechanical air; sustained directional airflow from clean to potentially contaminated; no reversal at the barrier under failure | Fan interlocks and standby |
| Airflow indicator | Visual device at the entry confirms direction; audible alarms considered | Gauge and alarm |
| Exhaust | Not recirculated to other building areas; dispersed away from occupied areas and intakes, or HEPA filtered | Outlet position or HEPA |
| Decontamination | Facility built so the whole laboratory can be decontaminated | Sealing and dampers |
| HEPA housings | Gas-tight isolation dampers, decontamination ports or bag-in/bag-out; leak-testable; certified at least annually | Housing type and access |
| Verification | Design and operating parameters verified and documented before use; tested annually or after major change | Commissioning plan |
Other facility items
The list also asks that vacuum lines in use be protected by liquid disinfectant traps and in-line HEPA filters, that designers consider ways to decontaminate large equipment before it leaves the room, and that communication and emergency access or egress be planned.
Enhancements a risk assessment may add
The BMBL lists optional enhancements: an anteroom for clean storage with dress-in and shower-out capability, gas-tight dampers to isolate the laboratory, final HEPA filtration of exhaust, effluent decontamination, containment of piped services and advanced access control. The owner decides which apply.
Questions for the air plant designer
The BMBL gives no pressure number in its BSL-3 list, only directional airflow that must not reverse. Ask the designer what pressure and air change values the national code requires, and how the system behaves when a supply or exhaust fan stops.
Ask where exhaust discharges, whether HEPA filters sit close to the room to shorten contaminated duct, and how each filter housing is leak-tested in place.
Testing and handover
Ask for records of directional airflow, alarm function, HEPA housing leak tests and damper tests. UK HTM 03-01 refers designers of containment level 3 rooms to the MRC's Standards for containment level 3 facilities, a national example of a separate rule set.
BSL-3 is separate from ISO 14644-1 class. A containment room may also need a particle class if the work includes clean processing. State both.
Who approves the room
Any approval comes from the authority your country sets, and it applies to the owner's laboratory, not the contractor. The owner's biosafety officer signs off the risk assessment. To have a room list reviewed, send it to ZY Cleanroom, a cleanroom design-and-build contractor with its own panel factory in Tianjin, China.
Questions
Common questions
Does BSL-3 require HEPA-filtered exhaust?
The BMBL accepts exhaust dispersed away from occupied areas and intakes, or HEPA filtered. Final HEPA filtration is a listed enhancement. National rules may require it.
Does the BMBL give a room pressure?
Not in its BSL-3 list. It asks for directional airflow that does not reverse under failure. National codes give values.
Can I build a BSL-3 laboratory in an existing building?
Possibly, if surfaces, doors and vents can be sealed and the whole room decontaminated. Survey the shell first.
Is BSL-3 the same as an ISO class?
No. Biosafety level concerns containment. ISO 14644-1 class concerns particle count.
